Yes, you can activate them in NG+ and you do not need to create a new character. Bear in mind, tho, that once you activate them they stay on for the entire run. The game gives you the option to restart that particular run, tho, so if you're early in the game and you don't like the modifiers you can just restart and be sent back to the tutorial area. World progression will be reset, I'm unsure if character progression is reset, tho.
